Sarcoidosis is a systemic granulomatous inflammatory disease affecting 150,000-200,000 Americans, characterized by CD4+ T-cell and macrophage activation forming non-caseating granulomas in lungs, lymph nodes, skin, eyes, heart, and nervous system. Sarcoidosis is driven by Th1 IL-12/IFN-gamma dysregulation and myeloid antigen-presenting cell overactivation, often with hypercalcemia from granuloma 1-alpha-hydroxylase.
