Mixed connective tissue disease (MCTD) is an overlap autoimmune syndrome combining features of lupus, scleroderma, polymyositis, and rheumatoid arthritis, defined by anti-U1 RNP antibodies. MCTD is driven by B-cell dysregulation, TGF-beta fibrosis, Raynaud's vasospasm, and multi-organ inflammation.
